Hepatoprotective Effects of Willow (Salix koreensis ANDERSS.) Branch Extracts against Cytotoxicity Induced by tert-Butyl Hydroperoxide in Human Hepatoma Cells

Abstract
The purpose of this study was to investigate the hepatoprotective effects of willow (Salix koreensis ANDERSS.) branch extracts (WBE) against tert-butyl hydroperoxide (t-BHP)-induced cytotoxicity in human hepatocarcinoma HepG2 cells. WBE were prepared using an ultrasonication method at a frequency of 132 kHz with 70% ethanol. Cell viability, activities of alanine transferase (ALT) and aspartate transferase (AST), malondialdehyde (MDA) levels and glutathione (GSH) levels were measured to evaluate the extent of cell damage. In t-BHP-induced toxicity, WBE treatment increased the viability of the HepG2 cells in a dose-dependent manner (P<0.05). WBE treatment significantly decreased the activities of ALT and AST as well as reduced MDA levels in HepG2 cells (P<0.05). Finally, WBE treatment significantly increased the GSH levels (P<0.05). These results suggest that WBE has significant hepatoprotective effects and could be a potential candidate for use in improving liver function.
